Some keys in Dead Island 2 can only be obtained by doing side quests. Either that, or some locked chests, boxes, or lockers are not available until you advanced into the story. How to Unlock Custodian’s Locker – Dead Island 2
First of all, you will need to complete the Missing Rainer quest. Without it, you will never be able to find the Custodian’s Locker Key.
Once you have completed the quest, you will then be able to pick up the Custodian’s Locker Key.
After getting the key, you can then head into this room and open the Custodian’s Locker with the key you picked up.
